Urine qPCR diagnoses over the entire clinical-immunological spectrum of human Leishmania (L.) infantum chagasi-infections in the Brazilian Amazon

The clinical-immunological spectrum of human Leishmania (L.) infantum chagasi-infections in the Brazilian Amazon has been defined using DTH/IFAT-IgG immune assays and clinical statuses infected individuals, revealing five profiles: three asymptomatic [Asymptomatic Infection (AI), Subclinical Resistant (SRI), Indeterminate Initial (III)], two symptomatic profiles [Subclinical Oligosymptomatic (SOI) Symptomatic (SI = American visceral leishmaniasis/AVL)]. We evaluated diagnostic potential urine qPCR over entire infection. Resine Instagene Matrix® was used for DNA extraction from urinary sediment, with amplification carried out SYBR® Green Taq RV1 RV2 primers. examined samples 151 individuals an endemic area AVL Pará State Amazon, including: 91 (60.3%) diagnoses previous infections [13 (14.3%) sharing AI profile, 13 SRI 43 (47.2%) III, 12 (13.2%) SI (treated AVL), 10 (11%) (untreated AVL)]; sixty (39.7%) were DTH(?)/IFAT-IgG(?) (the uninfected group). positive 61.5% both profiles, 65% III 50% treated AVL, 100% untreated 6.7% group.
